Chloramine-T and phenol red react to form a slightly colored reagent stream. A 70 cm mixing coil premixes the reagents to establish a stable baseline. A 200 μL sample is injected into the carrier stream, which merges with these color reagents. The bromide ions (Br–) are oxidized by the chloramine-T to Br2 (aq). The Br2 formed then readily brominates the phenol red. If the reaction is buffered at pH 4.5 to 4.7, the color of the brominated product will range from reddish to violet, depending on the bromide concentration. The absorbance, measured at 590 nm, is directly proportional to the concentration of the bromide in the sample.
